The growth and the toxin (i.e. hemolysin) producing capacity of Corynebacterium pyogenes were studied in monocultures and in co-cultures with 1 or more of the organisms frequently accompanying it in summer mastitis in cattle (Peptococcus indolicus, Stuart-Schwan cocci, Bacteroides melaninogenicus subsp. levii, Fusobacterium necrophorum and Streptococcus dysgalactiae) or with organisms seldom associated with summer mastitis (Streptococcus uberis, Streptococcus agalactiae, non-toxic staphylococci and Escherichia coli).Pc. indolicus, and to some extent also Stuart-Schwan cocci, stimulated the growth as well as the hemolysin producing capacity of Gb. pyogenes (Table 1) while Str. dysgalactiae, Str. uberis, Str. agalactiae, E. coli and the majority of the staphylococci reduced these activities. Most F. necrophorum strains stimulated the growth, but not the hemolytic activity. With B. melaninogenicus the results were inconclusive.The effect of Pc. indolicus appeared to be associated with the production of a filterable factor (Tables 2 and 3).Mouse toxicity and hemolytic activity of culture filtrates were closely correlated (Table 4).  相似文献

The study comprises 136 strains of Corynebacterium pyogenes originating from cattle (105), swine (20), sheep (1), and insects (10). For comparison 2 strains of human origin and 1 strain of Gorynebacterium hemolyticum were examined.One of the bovine strains was atypical, being gelatinase-negative, otherwise the strains of Cb. pyogenes were found to be biochemically identical apart from minor deviations in fermentation patterns (Table 1). Neither were antigenic differences demonstrated (gel diffusion analyses, Figs. 1 and 2).Both of the human strains agreed biochemically with Cb. pyogenes (Table 1). By gel diffusion cross analyses one of them was found to be identical with Cb. pyogenes, the other not, though anti-genically related to it (Fig. 2).Gb. hemolyticum deviated biochemically as well as serologically from Gb. pyogenes, but the 2 organisms shared antigenic determinants (Fig. 3).  相似文献

为探明贵州省某猪场引起猪体表脓肿的原因,本研究对该猪场脓肿部位的脓汁进行细菌分离培养,并对分离所得的细菌进行革兰氏染色镜检、生化试验、药敏试验、16S rDNA序列分子分析及动物感染试验。结果显示,从脓汁中成功分离到了3株菌落形态不一的菌株,分别为金黄色葡萄球菌、化脓隐秘杆菌、停乳链球菌类马亚种,根据分离地点和时间将其分别命名为GZGP2018-1、GZGP2018-2和GZGP2018-3;GZGP2018-1菌株与NCBI上金黄色葡萄球菌的同源性高达99.9%,GZGP2018-2菌株与NCBI上化脓隐秘杆菌的同源性高达99.9%,GZGP2018-3菌株与NCBI上停乳链球菌类马亚种的同源性高达100%;3株分离菌株对头孢拉定、环丙沙星、磺胺间甲氧嘧啶和氟苯尼考较敏感,对青霉素类药物和红霉素耐药;3株分离菌株对试验小鼠均具有致死性。本研究为该猪场猪体表脓肿的发病原因、实验室诊断方法及日常防控提供了理论参考。  相似文献

通过制备兔抗化脓隐秘杆菌(Trueperella pyogenes)高免血清,筛选固定方法、封闭液、洗涤液、抗体稀释度、抗体孵育时间等应用条件,建立一种检测化脓隐秘杆菌的间接免疫荧光方法。结果表明:用冷丙酮作固定剂,10%山羊血清作封闭液,0.05%Tween–20的0.01 mol/L pH 7.4 PBS(磷酸缓冲盐溶液)作洗涤液,兔抗化脓隐秘杆菌高免血清(一抗)1∶50稀释,37℃作用2 h,羊抗兔FITC(二抗)1∶100稀释,37℃作用2 h,草酸铵结晶紫衬染菌体3 min,对纯培养物化脓隐秘杆菌检测,可见清晰的特异性荧光,能区分溶血隐秘杆菌、伪结核棒状杆菌、金黄色葡萄球菌、大肠埃希菌、沙门氏菌、猪链球菌;在人工感染化脓隐秘杆菌死亡小鼠的心脏、肝脏、脾脏、肺脏、肾脏及发病山羊临床病料中均能检出化脓隐秘杆菌。  相似文献

为了解引起山羊皮下脓肿的主要病原,本研究从四川乐至县、金堂县和蓬溪县3个山羊养殖场无菌采集患皮下脓肿病羊脓液样本19份,通过细菌分离培养、生化试验及特异性PCR方法对其病原进行检测和鉴定,并通过药敏试验测定分离病原的药物敏感性。结果显示,PCR扩增法共检测出病原菌19株,其中伪结核棒状杆菌、金黄色葡萄球菌及化脓隐秘杆菌分离率分别为57.9%(11/19)、26.3%(5/19)及15.8%(3/19);采用细菌分离培养法分离到3种形态不同的病原菌共计17株,其中革兰氏阳性短杆菌经鉴定为伪结核棒状杆菌,分离率57.9%(11/19),革兰氏阳性球菌经鉴定为金黄色葡萄球菌,分离率为21.1%(4/19),革兰氏阳性杆菌经鉴定为化脓隐秘杆菌,分离率为10.5%(2/19),PCR鉴定较细菌分离培养法灵敏度更高。3种病原菌对丁胺卡那、氟苯尼考等多种药物敏感。结果表明,伪结核棒状杆菌是引起该地区山羊皮下脓肿的主要病原,金黄色葡萄球菌和化脓隐秘杆菌同样可引起山羊皮下脓肿,特异性PCR方法较细菌分离培养更为高效、准确。本研究结果可为后续进一步分析其生物学特性,以及四川地区山羊皮下脓肿的综合性防制提供依据。  相似文献

A total of 1050 samples from apparently healthy cattle were examined bacteriologically with special regard to Pc. indolicus and Cb. pyogenes.Pc. indolicus was found in 58 % of 130 samples from tonsils (slaughterhouse material), in 23 % of 620 samples from the vagina of cows, in 22 % of 100 samples from the vagina of calves and heifers, in 5 % of 100 samples from the conjunctival sac of cows, and in 10 % of 100 samples from the nasal cavity of cows (Table 1). Cb. pyogenes was found in 51 %, 17 %, 19 %, 8 %, and 6 %, respectively. Both organisms were found in each of 9 herds examined, though with varying frequency (Tables 2, 3, and 4).Altogether Pc, indolicus was found in 254 (24 %) and Cb. pyogenes in 205 (20 %) of the samples examined (Table 1). In 127 samples both organisms were present. Eleven of the strains of Pc. indolicus were β-hemolytic, the rest non-hemolytic.By gel diffusion analysis the strains of Pc. indolicus as well as those of Gb. pyogenes could be identified with strains originating from pathological conditions in cattle. With Serotype B occurring most frequently, usually two or three different types of Pc. indolicus were found in each of the herds examined (Tables 5, 6, and 7).The investigation has shown that Pc. indolicus is widespread among healthy cattle, and given evidence to suggest that Pc. indolicus and Cb. pyogenes are natural cohabitants.  相似文献

Abstract

CASE HISTORY: A 2-year-old Hereford bull was lame for one week before becoming recumbent.

CLINICAL AND PATHOLOGICAL FINDINGS: The scrotum and ventral perineal region were cold and blackened caudally. The semimembranosus and semitendinosus muscles were firm on palpation. The bull was subject to euthanasia, and necropsy revealed that the skin and S/C tissues of the caudal half of the scrotum were grey and necrotic. The caudal and distal aspects of the semimembranosus and semitendinosus muscles were grey and necrotic to a depth of approximately 15 cm, and these changes appeared to track along fascial planes. The tissue had an offensive smell, and large amounts of flocculent, watery, brown fluid and some gas were present. Histology of affected muscle and S/C tissues revealed coagulative necrosis, with oedema and large numbers of bacteria that were predominantly Gram-positive rods. Adjacent blood vessels contained thrombi while the epidermis overlying the affected areas appeared diffusely necrotic, suggesting infarction. Culture of the fluid yielded a pure growth of Arcanobacterium spp., which was identified as Arcanobacterium haemolyticum, using an API Coryne biochemical test strip.

DIAGNOSIS: Necrotising fasciitis and myositis due to Arcanobacterium haemolyticum.

CLINICAL RELEVANCE: Arcanobacterium haemolyticum has not previously been reported as a cause of necrotising fasciitis in any species. Necrotising fasciitis is probably an under-reported condition in cattle due to its clinical similarity to clostridial disease.  相似文献

从吉林省某猪场1例病死猪肺脏内分离到1株革兰氏阳性杆菌,该菌接种于TSA(含5%胎牛血清)平板,培养18h长出肉眼可见的菌落,而在巧克力平板上生长缓慢。对该菌株的16SrRNA基因进行PCR扩增并进行序列测定,测序结果经Blast分析与化脓隐秘杆菌的核苷酸序列同源性为99%,经过进一步糖、醇发酵试验及触酶试验等一系列生化试验,结果与测序结果相符,将其命名为AP4。该菌对供试小鼠具有致病性。药敏试验结果显示,该菌对氯霉素、氟苯尼考、阿米卡星及庆大霉素敏感,对红霉素、阿奇霉素、环丙沙星、强力霉素、阿莫西林及磺胺间甲氧嘧啶耐药。  相似文献
